RENAL FACTORS AFFECTING HEPATIC CHOLESTEROL SYNTHESIS

Abstract

Sterols are present in a ll ce lls containing double-layered subcellular membranes (1), probably serving as structural units in these membranes. Since the development of d is tin c t subcellular membranous structures permitted compartmentalization w ithin individual c e lls , larger and more complex ce lls were able to evolve. The a b ility to synthesize sterols is thus a key step in the evolution of the lower lif e forms (procaryocytes) to the plants and animals (eucaryocytes).Cholesterol is the major sterol present in mammalian tissues, but the amount present varies from tissue to tissue and from species to species (2). Cholesterol has been investigated extensively as a possible carcinogen w ithout the establishment o f an e tio lo g ic relationship (2, 3). A great deal o f in te re s t has prevailed in re la tin g cholesterol to atherosclerosis. The observation th a t cholesterol is a constituent o f the atherosclerotic plaque is over a century old (3). Early studies showed th a t the cholesterol content o f atherosclerotic aortas was greater than th a t o f normal aortas, that both free and e s te rifie d cholesterol were present, and th a t the ra tio o f free to e s te rifie d cholesterol approximated that found in blood. Other evidence that cholesterol is implicated in atherogenesis is the production of atherosclerosis by the feeding of cholesterol to susceptible animals. Further confirmation o f this re la tio n ­ ship is provided by the observation that human atherosclerosis is prominent in diseases accompanied by prolonged hypercholesterolemia.
