Application of A3 cytoplasm in sorghum

Roderick Devin Lee, University of Nebraska - Lincoln

Abstract

Prior to extensive evaluation in hybrid combination, a male-sterile version of a sorghum female parent must first be developed. Being able to identify the best combining female parents prior to or during the initial stages of male-sterilization would be advantageous. This would allow a sorghum breeder to be more effective and efficiently work the female germplasm pool. The following research was initiated to determine the feasibility of using A3 male-sterile lines as testers to evaluate early generation B-lines of the A1 CMS system (the CMS system used to produce nearly all hybrid sorghum seed grown commercially) in hybrid combination. B-line per se performance was also evaluated to compare the effectiveness of selecting for maturity, height, and grain yield in A3 hybrid combinations. The relationship between ms3 and reciprocal A3 hybrid combinations involving early generation B-lines was sufficient for advocating the use of A3 testers for screening early generation B-line material. The majority of superior B-lines could be identified by ranking and selecting the top 50%. Selecting less than the highest ranking 40% is not recommended based on this research since fewer of the superior B-lines would be identified. However, even selecting the highest ranking 40%-50% grain yielding B-lines, a sorghum breeding program can be more effectively and efficiently managed since inferior potential females can be discarded early in the line development process. For days to bloom and plant height (highly heritable traits), selection based on per se performance or in hybrid combination were similar.
