Use of forcing solution techniques to study the correlation of plant growth regulators with bud dormancy, subsequent rooting, in vitro responses and protein expression of forced softwood growth of woody plant species

Abstract

Chestnut, lilac, privet, spirea, white weeping oak and 5-leaf aralia were tested. Optimum bud break was obtained by soaking the basal 1/3 of dormant stems either in 10% bleach solution or in 75% ethanol solution for 15 minutes. Bleach or ethanol soaks exerted negative effects on bud break for spirea, but hastened bud break for lilac and privet. GA$\sb{\it 3}$ and BA in the forcing solution increased percent bud break and promoted shoot elongation. GA$\sb{\it 3}$ also hastened bud break while BA delayed bud break. IBA in the forcing solution decreased percent bud break and delayed bud break. IAA and NAA included in forcing solution enhanced subsequent rooting of forced softwood cuttings for privet. GA$\sb{\it 3}$ was initially added to the basic forcing solution for only 1 day or less for bud break and shoot elongation, then GA$\sb{\it 3}$ was replaced by adding IBA or other auxins to fresh basic forcing solution to "prime" the candidate softwood cuttings in advance for rooting. Addition of IBA following initial use of GA$\sb{\it 3}$ in the forcing solution counteracted the undesirable effects of GA$\sb{\it 3}$ on rooting and stimulated rooting following the favorable effects of GA$\sb{\it 3}$ on bud break and shoot elongation. BA and TDZ in the forcing solution promoted subsequent in vitro shoot regeneration of forced softwood growth of Acanthopanax sieboldianus. The forcing solution techniques also reduced the time needed from the initiation of in vitro culture to potted plants by half for this recalcitrant species. In vitro shoot initiation of softwood shoot explants was also significantly promoted by including BA, TDZ or CPPU in the culture medium. GA$\sb{\it 3},$ BA and IBA in the forcing solution influenced protein expression of privet and spirea. GA$\sb{\it 3}$ at 100 mg/l suppressed the shoot elongation, but still enhanced total protein expression. No statistically significant difference was observed for total protein expression between longer shoots and shorter shoots for privet.
